Background
Cbl (named after Casitas B-lineage Lymphoma) is a mammalian gene encoding the protein CBL which is an E3 ubiquitin-protein ligase involved in cell signalling and protein ubiquitination. Mutations to this gene have been implicated in a number of human cancers, particularly acute myeloid leukaemia. CBL acts as an E3 ubiquitin-protein ligase, which accepts ubiquitin from specific E2 ubiquitin-conjugating enzymes, and then transfers it to substrates promoting their degradation by the proteasome. It can recognize activated receptor tyrosine kinases, including PDGFA, EGF and CSF1, and terminates signaling. CBL protein is also an adapter protein that functions as a negative regulator of many signaling pathways that start from receptors at the cell surface.
